When you reach B4 you will be confronted by the Vampire's henceman, the Werewolf. Defeating it will allow you access to the Innermost Area, where the head Vampire current resides. If you collected the Chalice from St. Agnes' grave in North Estamir, you can do large amounts of damage using it's "Holy Sparkle" attack.

Depending on wether or not the player has obtained the holy chalice, the Vampiress will have more dialogue and a difference in music. If the quest takes place early in the playthrough, without obtaining the holy chalice, "the Conflict" will play and the Vampiress' dialogue will be severely reduced to a single sentence. If instead the quest is done after the player obtains the holy chalice from the South Estamir graveyard, the Vampiress will have alot more dialogue and "Beat them Up" will play instead.
Depending on wether or not the player has finished an event about [[Saruin]], the Vampiress will have more dialogue and a difference in music. If the quest takes place early in the playthrough, before any mention of Saruin in the story, "the Conflict" will play and the Vampiress' dialogue will be severely reduced to a single sentence. If instead the quest is done after Saruin is introduced into the main plot, the Vampiress will have alot more dialogue and "Beat them Up" will play instead.

*In the SNES version, defeating the Vampiress in the Den will actually cause Frontier to be invaded again, even if the Jewel Beast is dead. This seems to happen only if you beat her after Jewel Beast.
